원리

Milliplex-Luminex 시스템은 Suspension Array Technology(SAT)를 실현한 새로운 array system입니다. 기존의 특정장소에 probe가 고정된 평판 타입의 시스템들은 각 장소에 고정되어 있는 probe가 무엇인지 표지하기 위해서 장소를 지정하는 위치 정보가 사용되지만, SAT에서는 식별 코드를 부여한 bead 표면에 probe를 고정해, 이 bead의 식별 코드를 읽어내는 것으로 표면에 고정되어 있는 probe를 알 수 있습니다.

Luminex 시스템에서는, 담체로서 직경 5. 6μm의 polystyrene제 Microbeads 사용해, 이 Microbeads를 여러가지 농도에 편성한 2색의 형광 색소로 염색해, 각 형광 색소의 함유량을 식별 코드로서 이용하고 있습니다. 구체적으로는, 각 형광 색소에 붙어, detection sensor로 확실히 구별 가능한 10 단계의 농도를 선정합니다. 그것을 100배 하여 100 종류의 Microbeads를 작성합니다. 색 마다 다른 물질을 고정한 Luminex beads 세트를 준비해, 이 beads 세트와 reporter 형광 물질로 표지 한 샘플을 튜브 중(안)에서 반응 시킵니다. ↓ 샘플 결합 후, fluorometry 에 의해 Luminex beads를 하나씩 흘리면서, 적색 레이저와 APD 센서로 beads의 직경과 색을 측정해, 녹색 레이저와 광전자증배관에서 샘플의 결합양을 측정합니다



Milliplex cytokine detection systems are sandwich immunoassays performed on unique fluorescent beads, which contain a precise distribution of two fluorescent dyes, and a specific cytokine capture antibody linked to its surface. The fluorescent bead sets are incubated with a biological sample. A biotinylated cytokine reporter antibody is then used to identify the captured cytokine bound to the specific fluorescent bead. The reaction is detected by adding a streptavidin-phycoerythrin conjugate to the reaction mixture and read in a Luminex 100 instrument.
