Accutase (Cell detachment solution)

• 세포에 부드럽고 mild하게 작용됩니다.
• primary culture를 위한 tissue dissociation에도 사용가능합니다.
• mammalian이나 bacteria 유래 제품을 이용하지 않아, 오염으로부터 안전합니다.
• Trypsin이 enzyme inhibitor나 washing등을 필요로 하는 반면, Accutase는 이런 중화과정이 필요없습니다.
• SmartPlastic이라는 제품과 사용하여 cell을 제거할때도 효과적입니다.

제공형태
• 100 ml, ready to use, frozen sterile liquid. 1X ACCUTASE enzymes in Dulbecco's PBS (0.2 g/L KCl, 0.2 g/L KH2PO4, 8 g/L NaCl, and 1.15 g/L Na2HPO4) containing 0.5 mM EDTA·4Na and 3 mg/L Phenol Red.

적용실험
• Accutase performs exceptionally well in detaching cells for: analysis of cell surface markers, virus growth assay, quiescence assays by serum starvation, transformation assays by oncogene transfection, neural crest cell migration assays, cell proliferation, cell haptotaxsis, tumor cell migration assays, routine cell passage, production scale-up (bioreactor), and flow cytometry.

1. 1.ACCUTASE를 실온에서 녹인다.
2. 2.PBS로 plate, flask등을 세척한다.
3. 3.75cm2 surface area당 10ml의 ACCUTASE를 dish나 flask에 넣는다.
4. 4.37˚C, CO2 incubator에서 5-10분 배양한다.
5. 5.Cell counting and passage
   *떼어낸 cell에 대해, 세척액이나 enzyme inhibitor등으로 처리할 필요가 없다.

실험결과

Cell Detachment:
DMEM+10% FBS에서 배양된 Human MG63 fibrosarcoma cell culture를 Accutase로 처리하였다.
cell detachment가 빠르게 이루어져 5분후에는 다수의 single cell suspension이 관찰되었고, 세포의 viability도 높았다.
Accutase 처리 45분후에도 97 ± 3%의 높은율의 세포생존율을 확인할수 있었다.

Accutase 처리후에 plate로부터 세포가 모두 탈착되었음을 보여준다.
