Plant Direct PCR Kit

• DNA를 뽑지않고 plant leaf으로 바로 PCR을 수행하므로, 시간이 단축되며 간편합니다.
• Proteinase K를 사용할 필요가 없습니다.
• Harris Micro punch를 이용하여 정교하게 0.5mm의 leaf를 얻을 경우에는 lysis 과정없이 바로 PCR template로 사용할 수 있습니다.
• 칼등으로 조각을 내어 leaf를 얻을 경우에는 85°C 10분의 lysis step을 처리후, 추출액 1ul를 갖고 바로 PCR을 진행합니다.
• plant tissue를 갈거나, 여러 버퍼 및 DNA column을 이용하는 전통적인 방법에 비해 조작이 매우 간단하고 쉽습니다.
• 조작을 최소화함으로써 오염의 위험과 번거로움을 줄여줍니다.
• 실험과정이 단순합니다.
  1. 1) Plant leaf을 자른다.
        (Harris Micro punch로 0.5mm를 얻을 수 있는 경우에는 2,3번 과정없이 바로 PCR 수행한다.)
  2. 2) 함께 제공되는 Lysis buffer에 넣고 85°C에서 10분간 incubation 한다.
  3. 3) Vortex나 spin down
  4. 4) 상층액 1ul를 떠서 바로 PCR을 수행한다.

사용예

Direct PCR from Arabidopsis leaf:

PCR was performed with 1 ul Arabidopsis leaf lysate using the 1X Plant Lysis Buffer in 20 ul PCR. And, pieces of Arabidopsis leaf with 2 mm Harris Punch were placed directly into 20 ul PCR.
Lane 1, 6 and 11: negative control
Lane 2, 7 and 12: 1uL lysate
Lane 3, 8 and 13: one piece
Lane 4, 9 and 14: two pieces
Lane 5, 10 and 15: three pieces

Direct PCR from Plant leaves:

PCR was performed with 1 ul Nicotiana and Capsisum leaf lysate using the 1X Plant Lysis Buffer in 20 ul PCR. And, pieces of Nicotiana and Capsisum leaf with 2 mm Harris Punch were placed directly into 20 ul PCR.
Lane 1 and 5: 1uL lysate
Lane 2 and 6: one piece
Lane 3 and 7: two pieces
Lane 4 and 8: three pieces
