프로토콜

세척을 하지 않는 homogeneous한 비방사선 assay로, 간단한 2단계 프로토콜을 통해 90분 이내에 결과를 제공합니다. ADP가 PEP와의 반응에서 과산화수소(H2O2)를 생성하고, 이것이 peroxidase하에서 ADHP (형광염료 전구체)와 결합하면 형광 활성 resorufin dye를 생성합니다.  

HitHunter Kinase Activity Assay Kits assay principle. The assay principle is based on the ADP Hunter Plus detection method and allows for the measurement of ADP, a universal product of kinase activity. This homogeneous (no wash) assay is non-radioactive with a convenient gain-of-signal assay format and provides results in 90 minutes. This simple two-step assay uses a coupled enzyme reaction system that generates hydrogen peroxide (H2O2) from ADP. Hydrogen peroxide, when combined with ADHP (fluorescent dye precursor) in the presence of peroxidase, generates a fluorescent active resorufin dye. A reaction stop solution is also provided with the kit for added signal and background stabilization to allow for automation.
