특징 

• 세포나 조직 내에서 두 개의 관심 단백질이 서로 근접해 있는지(약 40nm 이내) 확인하고 정량하는 데 이용하는 시약입니다.
• 단백질 A, 단백질 B, A+B의 상호 작용 복합체, 3가지 상태를 동시에 정량할 수 있습니다.
• 두가지 형광 리포터를 이용하여 590nm, 664nm에서 확인합니다.
• Rolling Circle Amplification (RCA)라는 증폭 방법을 사용하여, 신호 감도가 높고 미세 상호작용도 검출할 수 있습니다.
• 120 cell assays (40 µl/assay) 혹은 60 tissue assays (80 µl/assay)가 가능합니다.

실험 과정

1. 1차 항체가 세포 내 단백질 A, B에 결합
2. 두 항체에 연결된 proximity probe가 DNA circle과 hybridization
3. DNA circle에 nicking (절단)
4. Tag oligo 삽입 및 ligation
5. RCA (rolling circle amplification) 수행 → 많은 복제된 DNA concatemer 생성
6. 형광 표지된 태그 oligo가 증폭된 DNA에 결합 → 형광 신호 검출
7. free 단백질의 경우에는 단일 형광 신호를, A‑B 복합체의 경우에는 두가지 형광이 겹쳐 나타남
