특징

 

  • 엑소좀을 포함한 세포외 소포체(EV)는 세포에서 방출되어 신체의 다른 부분에 있는 세포와 생물학적 정보를 전달합니다. 따라서 약물 전달 및 의료 진단 응용 분야에서 잠재적인 타겟으로서 연구되고 있습니다.
  • ExoBrite™ Staining Kit는 특히 flow cytometry를 이용한 유세포 분석에서 엑소좀 검출이 용이하도록 고안되었습니다. 기존의 일부 염료가 엑소좀과 유사한 응집체를 형성하여 분석에 혼란을 주기도 하지만, ExoBrite Stain은 백그라운드 문제가 전혀 없으므로 염색된 엑소좀만 정확하게 식별할 수 있고, 엑소좀 막의 분자에만 특이적으로 결합하여 밝은 형광을 띱니다.
  • 다른 membrane 염료들과 달리 polystyrene bead에 비특이적으로 결합하지 않으므로, 비드가 결합된 엑소좀을 염색하는 데에도 사용할 수 있습니다. 
  • 엑소좀은 하나 이상의 tetraspanin protein CD9, CD63, & CD81을 표적으로 하는 형광 항체를 사용하여 검출됩니다. ExoBrite stain은 multi-parameter 분석을 위해 항체 염색과 결합할 수 있습니다.

 SEC-purified EVs were stained with ExoBrite™ WGA EV Stains. MCF-7 derived EVs were stained with ExoBrite™ 410/450, ExoBrite™ 490/515, or ExoBrite™ 560/585. CHO derived EVs were stained with ExoBrite™ 640/660. 410/450 was detected in the Pacific Blue channel, 490/515 was detected in the FITC channel, 560/585 was detected in the PE channel, and 640/660 was detected in the APC channel.
